Molecular heterogeneity of cytosolic forms of estrogen receptors from human breast tumors.

چکیده
Molecular heterogeneity of estrogen receptors in cytosols from human breast carcinomas was demonstrated with regard to size and surface ionic charge. Following separation of the molecular forms of estrogen receptors into either the 8S or the 4S species by sedimentation on linear gradients of sucrose, diethylaminoethyl cellulose chromatography was used to fractionate the various com ponents in these receptors. Separation of the 8S species by diethylaminoethyl cellulose chromatography revealed a number of specific binding components, each eluting at a different ionic strength. The number of components as well as their relative specific estrogen-binding capacities were highly variable. At least five different species were identified on the basis of their elution at different ionic strengths. Estrogen-binding components eluted either in the wash buffer or at a KCI concentration of 0.02 to 0.05, 0.13 to 0.17, 0.18 to 0.21, or 0.3 M. The 4S form of the estrogen receptor separated into at least two compo nents, each eluting at a different ionic strength. Clinical correlations of objective remissions in breast cancer pa tients given hormonal therapy with the molecular species of estrogen receptors indicated that women with tumors containing the 8S form showed a greater likelihood of responding. Our findings with diethylaminoethyl cellulose chromatography suggest that the molecular composition of the 8S species of estrogen receptors from hormonally responsive tumors is different from that of certain unre sponsive neoplasms.

متن کاملThe hormonal milieu in primary breast cancer: A correlation between steroid receptors and serum estradiol, progesterone and prolactin
Background: The female breast is subjected to a lifetime of hormonal controls, whose effect is evident at the time of menarche and during the menstrual cycle, pregnancy and lactation. Studies have reported multiple risk factors for breast cancer, some of which are a reflection of hormonally mediated events. The steroid receptors are also served as prognostic factors for evaluating status of mal...
متن کاملCorrelation of Hormone Receptor Expression with Histologic Parameters in Benign and Malignant Breast Tumors
Background and Objective: Breast cancer is the commonest cancer of Indian women. Estrogen and Progesterone expression is seen in benign breast lesions and in breast carcinoma associated with good prognostic parameters and it correlates well with response to hormone therapy. Although a lot of studies have been conducted in the past on hormone receptor expression in breast cancer and few have cor...
متن کاملImaging features of estrogen-negative breast cancers: a correlation study with human epidermal growth factor type II overexpression
Background: Estrogen-negative breast cancers have different clinical course, prognostic features and treatment response in comparison to estrogen receptor-positive (ER-positive) breast cancers. Human epidermal growth factor receptor 2 (HER2) oncoprotein has found to have a pivotal role in natural cell growth and cell division and is suggested to be directly related to tumor invasiveness in brea...
متن کاملThe natural history of hormone receptor-positive breast cancer.
Abstract: Approximately 70% of human breast tumors express hormone receptors (HRs), comprising the estrogen receptor (ER) and/or progesterone receptor (PR).The ER is the primary transcription factor driving oncogenesis in HR-positive (HR+) breast cancers; it is both a target of, and predictor of response to, antiestrogen therapy. Unlike in other breast cancer subtypes, more than half of all dis...
